We resuspended the bead-bound nuclei in 100 μL antibody buffer (CUT&RUN Wash Buffer with 0.1% Triton X-100 (Sigma-Aldrich, 93443) and 2 mM EDTA (Invitrogen, 15575020)) and added 1 μg of antibody (IgG (Sigma-Aldrich, I8140) or H3K9me3 (Abcam, ab8898)).
